摘要:Objective: To investigate the mechanism of the effect of Moist Exposed Burn Therapy (MEBT) and Moist Exposed Burn Ointment (MEBO) on the spontaneous repair and healing of superficial third degree burn wound involving fatty layer. Method: A series of skin tissue samples were taken from deep burn wounds of 2 cases. Immunocytochemistry method, biotin-avidin DCS system, and indirect immunofluorescence technique were applied. Mouse anti-human keratin type 19 monoclonal antibody was used to detect regenerative epidermal stem cells in wound tissues. Results: Epidermal regenerative stem cells emerged at 24 hours post burn, and the number of epidermal regenerative stem cell increased on day 4 post burn. On days 7 and 14 post burn, the number of epidermal stem cells increased to the peak level. On days 21 and 28 post burn, it decreased and disappeared gradually as burn wound progressed to healing. 摘要:Moist Exposed Burn Ointment (MEBO?) is widely used topical agent applied on skin burn. This study investigated the effect of MEBO topical application on activation and proliferation of epidermal stem cells through the immunohistochemical localization of cytokeratin 19 (CK19) as a known marker expressed in epidermal stem cells. Biopsies from normal skin and burn wounds were taken from 21 patients with partial thickness burn 1, 4, 7, 14, 21, and 28 days after treatment with MEBO. Tissue sections were prepared for histological study and for CK19 immunohistochemical localization. In control skin, only few cells showed a positive CK19 immune-reaction. Burned skin showed necrosis of full thickness epidermis that extended to dermis. Gradual regeneration of skin accompanied with an enhancement in CK19 immune-reactivity was noted 4, 7, 14 and 21 days after treatment with MEBO. On day 28, a complete regeneration of skin was observed with a return of CK19 immune-reactivity to the basal pattern again. In conclusion, the enhancement of epidermal stem cell marker CK19 after treatment of partial thickness burn injuries with MEBO suggested the role of MEBO in promoting epidermal stem cell activation and proliferation during burn wound healing.
摘要:Dental Pulp Stem Cells (DPSCs) represent a new therapeutic alternative for treating superficial burn wounds. This study aimed to evaluate the effectiveness of DPSC-derived secretome on superficial burn wounds in an animal model. The study employed a post-test-only controlled group design using 15 rats with superficial burns. Rats were divided into a negative control group of 0.9% NaCl (K1), a positive control group of moist-exposed burn ointment or MEBO (K2), and a treatment group of DPSC-secretome (K3). The effectiveness of the DPSC-secretome was assessed by wound area percentage and histopathological observations on days 5, 14, and 21. DPSCsecretome significantly reduced the burn wound area (p<0.05). Histology data showed that the DPSC-secretome promoted re-epithelialization, fibroblasts, and collagen formation. Polymorphonuclear cells were also reduced after 21 days, indicating that the DPSCs-secretome has an anti-inflammatory effect. In addition, the DPSC-secretome and MEBO showed equal effects on angiogenesis after 14 days. The DPSC-secretome was more effective than MEBO on superficial dermal burns. This study showed that the DPSC secretome has therapeutic potential for burn wounds.
摘要:Secretome Dental Pulp Stem Cells have been proposed as a new alternative for wound burn. The aim of this study is to compare Secretome Dental Pulp Stem Cells and MEBO ointment against several fibroblasts and collagen in the healing process of superficial dermal burns. This study used a post-test only controlled group design by using 15 rats that were given superficial dermal burned wounds. Rats were divided into a negative control group of NaCl 0.9% (K1), a treatment group of MEBO (K2), and a treatment group of DPSCs-secretome (K3). Histopathology was done on the 5th, 14th, and 21st days post-induction wound burn. Network-prepared histopathology by colouring Messon Trichomes. Then, it was observed under a microscope with 200x magnification. Scoring in a way semi-qualitative for assessing fibroblasts and collagen. The total score of fibroblasts in the MEBO group was higher than the DPSCs-secretome group on day 5th. In comparison, the score for increasing the amount of collagen in the DPSCs-secretome group on days 5th and 14th was higher than the MEBO group. DPSCs-secretome has better effectiveness in increasing the amount of collagen but not better in increasing fibroblasts compared to MEBO.
