The effect of vitamin K on the wound healing process in rat skin achieved by common wound dressing agents

摘要:Background and objective: Vitamin K is a fat-soluble vitamin that plays an important role in the coagulation pathways of living organisms. A popular application of vitamin K has been used to help in the extravascular removal of blood from the skin. The aim of the present study was to test the wound healing effect of vitamin K and some common wound dressing agents available in Erbil city clinical settings such; the moist exposed burn ointment (MEBO) and the Cica silver spray to test their combination with vitamin K injections. Methods: Six groups of albino rats were used (6 rats in each group). Group M received Mebo ointment, Group Creceived Cica silver spray, Group K received Vitamin K injection, Group MK received Mebo ointment+vitamin K injection.Group CK received Cica silver spray +vitamin K Group, and Group N received no- treatment. The duration of the experiments was as 7, 14, and 21 days post wound surgery.The percentage of wound contraction was measured, and the blood serum was collected to test the level of transforming growth factor β (TGFβ) and platelet-derived growth factor (PDGF). Results: Data presented in our study showed that the best wound contraction percentage(99%) was obtained by the MK treated rats compared to the control group. Similar results were obtained fromTGFβ and PDGF data in which MK group showed a significant increase in the levels of these growth factors. Conclusion: The wound healing process is supposed to be due to an increase of TGFβ, PDGF, and enhanced fibroblast proliferation and neovascularization of tissues.

摘要:Objective: To investigate the mechanism of the effect of Moist Exposed Burn Therapy (MEBT) and Moist Exposed Burn Ointment (MEBO) on the spontaneous repair and healing of superficial third degree burn wound involving fatty layer. Method: A series of skin tissue samples were taken from deep burn wounds of 2 cases. Immunocytochemistry method, biotin-avidin DCS system, and indirect immunofluorescence technique were applied. Mouse anti-human keratin type 19 monoclonal antibody was used to detect regenerative epidermal stem cells in wound tissues. Results: Epidermal regenerative stem cells emerged at 24 hours post burn, and the number of epidermal regenerative stem cell increased on day 4 post burn. On days 7 and 14 post burn, the number of epidermal stem cells increased to the peak level. On days 21 and 28 post burn, it decreased and disappeared gradually as burn wound progressed to healing. 摘要:Factors involved in wound healing and their interdependence are not yet fully understood; nevertheless, new prospects for therapy to favor speedy and optimal healing are emerging. Reports about wound healing modulation by local application of simple and natural agents abound even in the recent literature, however, most are anecdotal and lack solid scientific evidence. We describe the effect of silver sulfadiazine and moist exposed burn ointment (MEBO), a recently described burn ointment of herbal origin, on mast cells and several wound healing cytokines (bFGF, IL-1, TGF-β, and NGF) in the rabbit experimental burn model. The results demonstrate that various inflammatory cells, growth factors and cytokines present in the wound bed may be modulated by application of local agents with drastic effects on their expression dynamics with characteristic temporal and spatial regulation and changes in the expression pattern. Such data are likely to be important for the development of novel strategies for wound healing since they shed some light on the potential formulations of temporally and combinatory optimized therapeutic regimens.

摘要:Moist exposed burn ointment (MEBO) is becoming increasingly popular in China as it shortens wound?healing time and reduces scar formation. However, its exact mechanism in mediating the wound?healing process is not yet clear. In the present study a total of 90?healthy adult male Wistar rats of specific-pathogen-free grade were divided equally into a control group, wound group, MEBO group, recombinant bovine basic fibroblast growth factor (rb?bFGF) group and sham operation group. Wound healing was observed from the extracted granulation tissues and recorded at three time points on 3, 7 and 14?days. Different levels of tumor necrosis factor?α (TNF?α) and interleukin?6 (IL?6) in tissue homogenate were detected using ELISA. Western blot analysis and quantitative PCR were used to detect the expression of nerve growth factor (NGF), substance P (SP) as?well?as tyrosine kinase?A (TrkA) receptor protein and the corresponding mRNA levels in granulation tissue. It was observed that the wound healing progressed faster in the MEBO and rb?bFGF groups compared with the wound group (P<0.01). TNF?α and IL?6 had an upward?downward trend at three time points, with the wound group demonstrating the most obvious increase (P<0.01). NGF and SP mRNA and protein levels in granulation tissue in MEBO, rb?bFGF and sham operation groups reached their highest levels on day?7 and then decreased on day?14. The expression level of TrkA was also measured simultaneously and its expression pattern was similar to that of NGF and SP. These results suggested that MEBO may promote nerve repair and accelerate wound healing through mediating the expression levels of NGF and SP, as?well?as TrkA.
